Cellular immunity to myelin basic protein in man and in animal model systems as measured by the macrophage migration inhibition test.

Lymphocytes from patients with neoplastic disease were tested for sensitization to encephalitogenic factor (EF) by the macrophage migration inhibition test. Sensitization to EF was demonstrated in 71% of patients with various forms of neoplastic disease. Sensitization to EF was also demonstrated for 31% of subjects with no evidence of neoplastic disease; these included patients with warts, chronic bronchitis and hernias. In contrast, healthy subjects showed no sensitization to myelin basic protein. These observations suggest that sensitization to EF may not be confined to patients with neoplastic disease. Lymphocytes from hamsters bearing a transplanted virus induced tumour were sensitized to EF prepared from both human and hamster brain. Sensitization was also seen in hamsters infected with influenza virus but not in animals with acute tubular necrosis produced by glycerol treatment. The development of an animal model system provides a method for the investigation of possible mechanisms of sensitization.

A quantitative study of alpha-1-antirypsin and alpha-2-macroglobulin in the sputum showed an increase in both during attacks of asthma and in patients with chronic obstructive bronchitis. The levels were much lower in asthma patients during remissions, in emphysema and in patients with chronic simple bronchitis, without associated bronchospasm. The parallel variations in levels of alpha-1-antitrypsin, albumin and transferrin in the sputum, contrasts with the course of secretory IgA and are in favour of a transudation process of alpha-1-antitrypsin across the respiratory mucosa.

Using affinity chromatography on concanavalin A (Con A) sepharose CL 6B, a carbohydrate-containing fraction was derived from Mycobacterium tuberculosis H37Rv sonicate. Surprisingly, the main component of Con A fraction was a protein having a molecular weight of a 30-kD range which is generally absent in the Con A-adsorbed fraction from the culture filtrate. ELISA by means of an antimycobacterial monoclonal antibody panel showed that the 30-kD range of Con A fraction contained antigen 85. It is suggested that the derived components antigen 85 are a glycosylated form of the proteins associated with the mycobacterial cell wall. The Con A fraction, antigen 85 (Department of Virology, Pasteur Institute, Brussels, Belgium), and PPD (Batch RT 45, Stattens Seruminstitute, Denmark) were used for ELISA determination of antimycobacterial antibody titers in the sera of 30 patients with pulmonary tuberculosis, 28 patients with nonspecific lung diseases (bronchitis and/or asthma, pneumonia), as well as in the sera of 12 healthy volunteers. The sensitivities were 46.42, 57.34, and 72.33% and the specificities were 36.97, 26.73, and 56.75% for Con A fraction, antigen 85, and PPD, respectively. The authors suggest that serodiagnostic properties of Con A fraction are extremely limited.

The macroscopic findings of 2385 autopsy protocolls of 70-102 year-old males and females were analysed for frequency and functional significance of diseases. 1. Severe coronary arteriosclerosis was found up to 60% of the cases, with increasing frequency in higher age groups. However decrease of acute myocardial necrosis was stated including the highest age group. 2. The mean values of heart weights decrease in the highest age groups. 3. While the frequency of arteriosclerosis in the cerebral arteries increases the frequency of ischemic lesions of the brain decreases. 4. Benign hyperplasia of the prostate is found in 85% of the cases. 5. 80% of the patients had severe substantial chronic pulmonary emphysema and acute tracheobronchitis. These pulmonary lesions are supposed to be the functionally most important disease in patients of old age.
