Ca2+ signals regulate many cellular functions, including hormone secretion. Agonist-specific Ca2+ signaling may arise from the differential mobilization of multiple Ca2+ stores. Although they act through the same receptor subtype, two gonadotropin-releasing hormones (sGnRH and cGnRH-II) generate quantifiably different Ca2+ signals in goldfish gonadotropes, suggesting that their Ca2+-dependent signaling cascades may differ. We combined electrophysiology, Ca2+ imaging, and radioimmunoassay detection of gonadotropin (GTH-II) secretion to determine the role of intracellular Ca2+ stores in GnRH-stimulated exocytosis. Our findings suggest that voltage-gated Ca2+ channels do not mediate acute GnRH-signaling. Instead, both sGnRH- and cGnRH-II-stimulated GTH-II releases are dependent on Ca2+ mobilized from TMB-8/CPA-sensitive compartments. However, sGnRH, but not cGnRH-II, utilizes intracellular stores sensitive to caffeine and xestospongin C. We also identified a homeostatic mechanism where reduced extracellular Ca2+ availability increase GTH-II release by mobilizing Ca2+ stores. Our results are the first to suggest that several classes of intracellular Ca2+ stores differentially participate in agonist signaling and homeostasis in gonadotropes.

The aim of this work was to study various transport phenomena in chitin gels obtained by N-acetylation of chitosan in a water-alcohol mixture. Three kinds of transport were investigated: the sorption of solutes interacting with chitin, the desorption of solutes without significant interaction with the polymer, and osmosis phenomena. In the case of interactive sorption, dyes having different chemical structures such as C.I. Acid Blue 74, C.I. Reactive Violet 5 or C.I. Direct Red 28 were tested. Sorptions of C.I. Acid Blue 74 and C.I. Reactive Violet 5 depend on the charge density of the polymer network and, as a consequence, on DA, pH and the dielectric constant of the media. This result reveals the importance of electrostatic interactions. On the other hand, the sorption of C.I. Direct Red 28 is mainly due to hydrophobic interactions and H-bonding, it is limited to the extreme surface of the gel. Concerning the non-interactive desorption, solutes of different steric hindrance such as PP vitamin, B1 vitamin and caffeine exhibit similar diffusion coefficients located within 3.7-5.6x10(-6) cm(2) s(-1). Finally, the osmotic behaviour of the gel immersed in a concentrated solution of gelatin allows us to multiply by 25 the concentration of chitin in the gel without any penetration of gelatin.

Effects of alprazolam, caffeine, and zolpidem in humans trained to discriminate triazolam from placebo.

This study compared the discriminative stimulus effects of zolpidem, a nonbenzodiazepine hypnotic, to benzodiazepines. Eight participants learned to discriminate triazolam (0.35 mg/70 kg) from placebo. The discriminative stimulus effects, self-reported subjective effects, and performance effects of triazolam (0.05-0.35 mg/70 kg), alprazolam (0.25-1.75 mg/70 kg), zolpidem (2.5-35 mg/70 kg) and caffeine (75-525 mg/70 kg) were assessed under two-response and novel-response drug discrimination procedures. Under the two-response procedure, triazolam, alprazolam and zolpidem fully substituted for triazolam and caffeine did not. Under the novel-response procedure, triazolam and alprazolam substituted for triazolam and zolpidem partially substituted for triazolam. Zolpidem, but not triazolam or alprazolam, also produced some novel responding. Caffeine produced both placebo-appropriate and novel responding. The self-reported effects of triazolam, alprazolam and zolpidem were similar. Overall, zolpidem produced similar, but not identical, effects as the benzodiazepines.
