Improvement of rice (Oryza sativa L.) seed oil quality through introduction of a soybean microsomal omega-3 fatty acid desaturase gene.

Microsomal omega-3 fatty acid desaturase is an essential enzyme in the production of the n-3 polyunsaturated fatty acid alpha-linolenic acid during the seed developing stage. We have constructed a chimeric gene consisting of a maize Ubi1-P-int and a soybean GmFAD3 cDNA, which was introduced into rice plants by Agrobacterium-mediated transformation. Ten transformants containing the chimeric gene were established and expression subsequently confirmed by Northern blotting. Furthermore, alpha-linolenic acid content of the T(1) seeds increased dramatically up to tenfold that of the control, and this phenotype was also stably inherited in the T(2) and T(3) progenies. These results demonstrate that the alpha-linolenic acid content of rice seed oil can easily be altered using the combination of a high-activity promoter and a GmFAD3 gene.

[Effect of ecological organic fertilizer on tomato bacterial wilt and soil microbial diversities]

The effects of ecological organic fertilizer (EOF) on tomato bacterial wilt and soil microbial community were studied through field trial and fatty acid methyl esters (FAMEs) analysis. The results showed that the incidence rate of tomato bacterial wilt was 100% in continuous cropped soil, and significantly reduced to 39% and 50% when applying EOF at 1.0 kg.m-2 and 0.5 kg.m-2, respectively. There was no significant difference between two fertilization treatments. The fatty acid fingerprints of soil microbial community were greatly changed, and the relative contents of cyc17:0 cited as biomarker for GN bacteria, c18:2 w6 as biomarker for fungi and c16:1 11c as biomarker for arbuscular mycorrhizal (AM) fungi all increased when applying EOF. The minimum distance of cluster analysis was 1.08 between control and treatments of applying ecological organic fertilizer, and 0.70 between different fertilization treatments. Applying EOF could adjust soil microbial community structure, benefit for soil beneficial microbes, and develop stability and disease suppression of soil ecological system, and hence, improve soil quality.

Impact of nisin producing culture and liposome-encapsulated nisin on ripening of Lactobacillus added-Cheddar cheese.

This study aimed to evaluate the effects of incorporating liposome-encapsulated nisin Z, nisin Z producing Lactococcus lactis ssp. lactis biovar. diacetylactis UL719, or Lactobacillus casei-casei L2A adjunct culture into cheese milk on textural, physicochemical and sensory attributes during ripening of Cheddar cheese. For this purpose, cheeses were made using a selected nisin tolerant cheese starter culture. Proteolysis, free fatty acid production, rheological parameters and hydrophilic/hydrophobic peptides evolution were monitored over 6 mo ripening. Sensory quality of cheeses was evaluated after 6 mo. Incorporating the nisin-producing strain into cheese starter culture increased proteolysis and lipolysis but did not significantly affect cheese rheology. Liposome-encapsulated nisin did not appear to affect cheese proteolysis, rheology and sensory characteristics. The nisinogenic strain increased the formation of both hydrophilic and hydrophobic peptides present in the cheese water extract. Sensory assessment indicated that acidic and bitter tastes were enhanced in the nisinogenic strain-containing cheese compared to control cheese. Incorporating Lb. casei and the nisinogenic culture into cheese produced a debittering effect and improved cheese flavor quality. Cheeses with added Lb. casei and liposome-encapsulated nisin Z exhibited the highest flavor intensity and were ranked first for sensory characteristics.

Effects of propylene glycol or fat drench on plasma metabolites, liver composition, and production of dairy cows during the periparturient period.

Forty-eight Holstein cows were used to determine the effects of short-term oral drenches of propylene glycol (PG) and Ca-soaps of palm oil fatty acids (fat) on plasma concentrations of key metabolites, liver composition, and milk production during the first 3 wk of lactation. Treatments (2 x 2 factorial arrangement) given orally once daily for the first 3 d postpartum were water (control), 500 ml of PG, 454 g of fat, or 500 ml of PG plus 454 g of fat. All treatments were administered as a total volume of 1.9 L. Administration of PG decreased concentrations of nonesterified fatty acids (NEFA) in plasma during the first 7 d and the first 21 d postpartum and tended to decrease concentrations of beta-hydroxybutyrate during the first 7 d postpartum. Concentrations of insulin in plasma were not affected by treatment. Administration of either PG or fat increased plasma glucose and liver glycogen concentrations compared to the control or PG plus fat treatments. Concentrations of triglycerides in liver were not affected by treatment. Administration of PG did not affect dry matter intake or milk yield and composition during the first 3 wk postpartum; however, cows drenched with fat tended to have lower dry matter intake and milk yield during the first 3 wk of lactation. Short-term drenching of PG effectively decreased NEFA concentrations in plasma during early lactation; however, data do not support administration of fat via drench to early lactation cows and concurrent administration of dietary fat appears to blunt the metabolic response of cows to PG.
