PIP: Fetal resorption caused by a single 10 IU sc injection of pregnant mare's serum gonadotropin (PMSG) on Day 5 in rats was reversed by 5 mg hydroxyprogesterone caproate sc on Days 4-15, and by 2 mg ovine prolactin sc on Days 4-15. Implanted blastocysts were counted on day 8 and rats were killed on Day 16 when weights of fetuses, placentae, ovaries, and corpor lutea (cl) were recorded. In PMSG-treated rats pregnancy was resorbed, ovaries were heavier with multiple antral follicles and lutein bodies, and individual CL were smaller. Both progesterone and prolactin maintained normal fetal, placental, ovarian, and CL weights and fetal survival. The results are attributed to estrogen-progesterone ratios.

High and very high dosages of the oral contraceptive lynestrenol: a three-month feeding study in female rats.

PIP: Lynestrenol, a progestagen used as an oral contraceptive, was tested in 105 Wistar female rats. Group A rats received .08 mg/kg/day for 3 months; Group B rats received .8 mg/kg/day for 3 months; Group C rats received 8 mg/kg/day for the 1st month, 16 mg/day for the 2nd month, and 32 mg/day for the 3rd month; and Group D rats served as controls without medication. Groups A and B were given the drug with their diet. Group C was dosed by esophageal tube. Some rats in each group died during the investigation. Group C rats consumed 10% less food than control rats and had a slower weight gain (p .001). Ratios of weights of liver, spleen, and kidneys to body weight were high in Group C rats. In Groups B and C rats, ovaries were small and corporalutea were rare and small. Vesicular ovarian follicles were mostly normal. Endometrial glands were less numerous and many were atrophic. It is concluded that lynestrenol was well tolerated by the rats for 3 months. An ovarian-blockade effect had been produced in the rats.

Effects of anterior hypothalamic deafferentation on LH, FSH, prolactin and sexual behavior in the ovariectomized ewe.

PIP: The effects of anterior hypothalamic deafferentation on luteinizing hormone (LH), follicle stimulating hormone (FSH), prolactin (PRL), and sexual behavior in the ovariectomized ewe was investigated. Response to injection of estradiol benzoate (50 mcg) was measured before and after surgery. In all 4 cases the estradiol-induced release of LH and PRL observed prior to surgery was eliminated after deafferentation. FSH and basal-PRL levels were also affected. After 3 months, 2 ewes showed partial recovery. Sexual behavior was eliminated in 2 cases. In the ewe, the anterior dorsal inputs from the preoptic anterior hypothalamus are required for the positive feedback of estrogens on LH and maintenance of FSH levels.

PIP: Prolactin is a polypetide hormone structurally similar to a growth hormone. It is secreted by lactotrophs in the anterior pituitary gland. The levels rise during pregnancy in response to estrogens formed by the feto-placental unit. Hyperprolactaemia in women causes a characteristic pattern of reproductive disturbance. The most severe form accounts for 15-20% of amennorhoea. Subnormal secretion of estradiol occurs because ovarian follicles do not mature. Symptoms are dyspareunia and dryness during intercourse. Hyperprolactinaemia in men suppresses gonadtropin secretion, and testosterone concentrations may be subnormal. Suppression of prolactin can be achieved by withdrawing drugs which provoke prolactin release, by correcting hypothyroidism and by direct treatment of a prolactin-secreting pituitary tumor. Bromocriptine suppresses prolactin secretion by direct action on the pituitary lactotrophs. It may be possible to avoid procedures such as radiation therapy and surgery in patients with small pituitary tumors who wish to become pregnant, but close cooperation of patients in special units is essential.
