In vitro effect of leptin on somatolactin release in the European sea bass (Dicentrarchus labrax): dependence on the reproductive status and interaction with NPY and GnRH.

The aim of the present work was to investigate the neuroendocrine control of pituitary somatolactin (SL) release using dispersed pituitary cell culture obtained from male European sea bass (Dicentrarchus labrax) at different stages of sexual development. The effect of mouse recombinant leptin, sea bream gonadotropin releasing-hormone (sbGnRH) and porcine neuropeptide Y (pNPY) and their potential interaction on the SL release were investigated. High doses of leptin (10(-8)-10(-6)M) were differentially effective in inducing SL release depending on the sexual developmental stage. Porcine NPY alone was not effective on basal SL release, but it dose-dependently (0.1 and 1 nM) enhanced SL release induced by leptin (10(-6) and 10(-8)M) in late pre-pubertal but not in post-pubertal stages. No effect of sbGnRH in association or not with leptin was observed on SL release. These findings are the first evidences that leptin and pNPY can play an important role in the neuroendocrine control of pars intermedia function and SL release in fish. In addition, the sensitivity of SL producing cells to leptin and NPY only in prepubertal and pubertal stages, provides the potential role of SL in the nutritional control of the onset of puberty.

The mechanisms for activating the hypothalamic-pituitary-adrenal (HPA) axis and the roles glucocorticoids play in the pathogenesis of chronic infectious disease are largely undefined. Using the LP-BM5 model of retrovirus-induced immunodeficiency, we found alterations in HPA axis function, manifested as an increase in circulating levels of adrenocorticotropic hormone and corticosterone, beginning after only 3 mo of infection. These changes occurred contemporaneously with a shift in the profile of circulating cytokines from a Th1-dominant (IFN-gamma) to Th2-dominant (IL-4, IL-10) phenotype. No significant changes in either circulating IL-1beta, IL-6, or TNF-alpha levels were observed in infected mice. Administering the N-methyl-D-aspartate receptor antagonist MK-801 to infected mice normalized plasma adrenocorticotropic hormone and corticosterone levels, indicating that glutamate was a major activator of the HPA axis. Moreover, MK-801 treatment of late-stage mice also reversed the type 1 to type 2 cytokine shift to a degree comparable or superior to treatment with the glucocorticoid receptor antagonist RU-486. These findings indicate that HPA axis activation during LP-BM5 retrovirus infection is mediated by the chronic hyperactivation of glutamatergic pathways in the hypothalamus. Through this mechanism, the degree of peripheral immunodeficiency observed in the late-stage disease is profoundly augmented.

In the course of producing monoclonal antibodies to turkey prolactin, three monoclonal antibodies to turkey chromogranin A (CgA) were also produced, apparently arising from minor contamination of the turkey prolactin immunogen with peptide fragments of CgA. The identity of the antigen recognized by these antibodies was established by tandem mass spectrometry de novo sequencing of seven tryptic peptides from a turkey pituitary protein purified by immunoaffinity chromatography. These peptides showed high homology with distinctly separate regions of mammalian and ostrich CgA, and in silico cloned chicken CgA sequences. Chromogranin A immunostaining patterns on Western blots and pituitary tissue sections differed from those of prolactin, growth hormone, or luteinizing hormone (LH). Dual-label fluorescent immunohistochemistry revealed that CgA was co-localized with LH in most avian gonadotrophs in young chickens and turkeys, but not in adult, laying birds. Conversely, CgA was found in a majority of somatotrophs in laying birds but was absent from somatotrophs in young, growing chickens and turkeys. Lactotrophs contained no detectable CgA immunoreactivity in the tissues studied. These results suggest that CgA may modulate hormone secretion by gonadotrophs and somatotrophs in a manner that differs between cell type with age or reproductive state.

Interference in radioimmunoassays (RIAs) was frequently encountered during endocrinological studies of the emu (Dromaius novaehollandiae). Interference was greatest when serum was cloudy or opaque. Such samples appeared seasonally, in spring and summer during the phase of fat deposition, and in the winter when females were laying. These poor quality samples did not allow accurate measurement by RIAs of several peptide hormones for a full year. To prepare them for assay, these sera were clarified using a polyethylene glycol (PEG) solution at a final concentration of 7.5%. This treatment was effective in most cases. After treatment with the PEG, recoveries of LH, glucagon, and prolactin were greater than 75% and that for insulin was 40%. Regardless of the level of recovery, there was a high correlation of assay results between non-opaque native and PEG-treated sera. Serum samples containing large amounts of interfering high molecular weight components, such as lipoproteins, can be clarified with PEG, enabling their accurate measurement by RIA.

Characterization of thyroid hormone receptor alpha and beta in the metamorphosing Japanese conger eel, Conger myriaster.

Two distinct TR alpha cDNA clones (TR alpha A and TR alpha B) were isolated from conger eel (Conger myriaster). The deduced amino acid sequences of the conger eel TR alphas showed higher homologies to the TR alphas of other vertebrates than to TR betas. Determination of TR mRNA in metamorphosing eels was performed using competitive RT-PCR. Of the two TR alpha mRNAs identified, TR alpha A mRNA expression was shown to be relatively higher than that of TR alpha B, and there was a peak in the expression of each during metamorphic climax. We hypothesize that both TR alphas play important roles in morphological differentiation during metamorphosis. The expression pattern of TR beta 1 mRNA was also higher during metamorphic climax and high levels of expression continued after metamorphosis. This suggests that TR beta 1 is the adult fish form which appears at high frequency after metamorphosis. It was also shown that TR beta 2 is highly expressed specifically in the brain and pituitary gland in larvae and juvenile forms including during metamorphosis and there was a peak in TR beta 2 mRNA in the elver after metamorphosis. Thus, we propose that TR beta 2 plays an important role in the regulation of the hypothalamic-pituitary-thyroid axis.
