Effects of vasoactive intestinal polypeptide (VIP) on lower esophageal sphincter in awake baboons: comparison with glucagon and secretin.

The effects of vasoactive intestinal polypeptide (VIP), glucagon, and secretin on lower esophageal sphincter pressure were investigated in awake baboons. The three hormones were compared with respect to effect on (1) resting lower esophageal sphincter pressure and (2) maximal stimulatory response to pentagastrin. VIP was shown to reduce resting and pentagastrin-stimulated lower esophageal sphincter pressure with significantly greater potency than either secretin or glucagon. For reduction of resting lower esophageal sphincter pressure, the potency ratio of VIP to secretin was 16:1 and of VIP to glucagon was 32:1 (P less than 0.05). For inhibition of pentagastrin-stimulated sphincter pressure, the potency ratio of VIP to secretin was 32:1 and of VIP to glucagon was 64:1 (P less than 0.02). This demonstration of significantly increased potency of VIP over known inhibitory hormones strengthens the suggestion that VIP may have a physiologic role in the control of lower esophageal sphincter function.

The dynamics of testosterone and dihydrotestosterone metabolism in the adult male rhesus monkey.

MCRs of testosterone (T) and 5 alpha-dihydrotestosterone (DHT) were determined in the morning and evening in four chronically chaired adult rhesus monkeys (Macaca mulatta) by means of steady state infusion of [4-14C]testosterone and [1,2-3H]dihydrotestosterone. Morning and evening MCRs were comparable for both T and DHT, averaging 169 and 212 ml/min for T and 93 and 108 ml/min for DHT, respectively, indicating that temporal differences in blood steroid concentrations are the result of changes in glandular secretion and/or synthesis and not differences in MCRs. Blood transfer factors for the conversion of T to DHT were also similar at these two times (0.024 and 0.022). MCRs were determined in one monkey by the single injection of [14C]T and [3H]DHT. From the rates at which the hormones disappeared from the circulation, MCRT and MCRDHT were calculated to be 172 ml/min for T and 92.2 ml/min for DHT. The inner volumes of hormone distribution were 4.04 liters for T and 3.67 liters for DHT.

Effects of 1,25-dihydroxyvitamin D3 on the distribution of androgen and vitamin D receptors in human prostate neonatal epithelial cells.

Although many studies have examined the mechanisms of 1,25-dihydroxyvitamin D(3) (calcitriol or 1,25 D) action in different prostate cancer cell lines, little is known regarding the influence of this steroid on the normal prostate. The presence of both VDR and AR in normal prostatic tissues raises the distinct possibility of an important role for this hormone in the normal gland. In order to ascertain the possible role of 1,25 D on both AR and VDR in the normal prostate, the effects of calcitriol and dihydrotestosterone (DHT) on the normal human neonatal prostatic epithelial cell line, 267B-1, were examined. These studies were approached by focusing on how 1,25 D in the presence or absence of DHT affects the distribution of AR and VDR in the cytoplasmic and nuclear compartments of the cells in terms of their protein levels and DNA binding activities. Immunoblot analyses show that 1,25 D increases the AR protein level in both the cytoplasmic and nuclear fractions but not the VDR protein level. On the other hand, the gel shift assays demonstrate that 1,25 D increases both the AR- and VDR-DNA binding activities in the nuclear fraction, whereas there is no increase in DNA binding activities in the cytoplasmic fraction. Addition of DHT along with 1,25 D does not affect the DNA binding activities of both AR and VDR. Overall, these studies suggest that 1,25 D actions on the normal prostate cells may be mediated independently through AR and VDR, respectively. 2002 Wiley-Liss, Inc.

BACKGROUND: The influence of sex steroids upon brain development has been suggested to mediate sex differences in developmental psychopathology. The ratio of the length of index finger or second finger to the ring finger or fourth finger (the 2D:4D ratio) appears to be a marker of early sex hormone exposure, with low 2D:4D associated with high prenatal testosterone and high 2D:4D associated with high prenatal oestrogen. This relationship allows a non-invasive measure of the long-term influence of prenatal sex steroids. Behaviours such as hyperactivity and poor social cognition are common in preschoolers. An association between 2D:4D and these possible precursors of psychopathology would be most readily identified in this group. AIM: To identify relationships between 2D:4D ratio and behaviours in preschool children which constitute possible precursors of developmental psychopathology. STUDY DESIGN: Population survey. METHOD: The 2D:4D ratio was measured in a group of preschool children and behavioural questionnaires were given to parents and teachers. RESULTS: Sex differences in behaviours were small, whilst correlations with 2D:4D were strong. Low 2D:4D was related to hyperactivity and poor social cognitive function in girls, and high 2D:4D with emotional symptoms in boys. CONCLUSIONS: We suggest that during early brain development androgens increase the probability of hyperactivity and poor social cognition in girls. Early oestrogens increase the probability of emotional problems in boys.

Specific binding of 20-hydroxyecdysone to nuclei of imaginal discs of Drosophila melanogaster.

Specific binding of the insect steroid hormone 20-hydroxyecdysone to imaginal discs of Drosophila melanogaster has been investigated. Evidence is presented showing that most of the specific binding is located in the nuclear fraction at the time changes in gene function are observed. Nuclear binding is high affinity, analog specific, apparently saturable, and unaffected by inhibitors of RNA and protein synthesis. The association kinetics of nuclear binding are very similar to those of specific binding in whole cells. Specific binding to whole discs and to disc nuclei is temperature-dependent, but equal levels of nuclear binding are achieved after 1 h at 25 degrees C and 8 h at 0-4 degrees C. There is little or no lag in the nuclear location of specific binding at either temperature. The biochemical properties of the specific nuclear binding are consistent with the involvement of these sites in the hormone detection and response system mediating imaginal disc morphogenesis.
