Inhibition of lung metastasis of B16 melanoma cells exposed to blue light in mice.

The effects of blue light on B16 melanoma cells and on the metastasis of these cells to the lungs were investigated in mice. The exposure of B16 melanoma cells to blue light in two 20-min sessions resulted in marked suppression of cell growth measured at 7 days after exposure. When these cells were harvested, re-inoculated into medium and incubated for a further 7 days, their growth activity returned to almost the same level as that of cultured cells from the non-exposure control group. The melanoma cells harvested after 7 days of incubation were injected intravenously into mice. In the non-exposure group, black nodules developed on the lung surface and the nodules increased in size over time. In the blue-light-exposure group, the development of such black nodules on the lung surface was delayed, and the nodules were smaller. Histopathological examination revealed that blue light suppressed the growth of metastatic tumor cells, and no increase in the number of melanin-containing cells or atypical cells was induced in the metastatic lesions. These results suggest that blue light suppresses the metastasis of B16 melanoma cells.

Acral lentiginous melanoma: an immunohistochemical study of 20 cases.

BACKGROUND: Though acral lentiginous melanoma (ALM) is a major type of malignant melanoma, no immunohistochemical study on this type of melanoma has been reported. OBJECTIVE: The purpose of this study is to analysis the immunohistochemical findings of ALM using routinely used immune markers. METHODS: An immunohistochemical study was performed on paraffin sections of 20 ALMs using S-100 protein, HMB-45, MART-1, vimentin, epithelial membrane antigen (EMA) and CAM 5.2. RESULTS: S-100 protein (95%) was found to be a more sensitive marker than either HMB-45 (80%) or MART-1 (70%) for recognizing ALM. Melanin bleaching was useful for recognizing heavily pigmented ALM using both S-100 protein and HMB-45. The intensity of HMB-45 correlated well with the melanin content. However, there was no significant correlation between the intensity of S-100 protein and the melanin content. One and two out of 20 cases stained focally with EMA and CAM5.2, respectively, but these cases stained also with HMB-45 and/or S-100 protein. CONCLUSIONS: S-100 protein and HMB-45 were relatively sensitive markers for recognizing ALM. Despite the occasional positivity for the epithelial markers in ALM, all epithelial marker-positive cases stained also with HMB-45 and/or S-100 protein. Therefore, we recommend that the panel of antibodies used for recognizing ALM should contain at least S-100 protein and HMB-45.

Differential effects on melanocyte growth and melanization of low vs. high calcium keratinocyte-conditioned medium.

Epidermal keratinocytes secrete several growth factors that stimulate melanocyte proliferation and melanin pigment synthesis in vitro. As the epidermis is formed of two distinct layers, i.e. basal cell layer and suprabasal cell layers, and both are functionally and biologically different compartments, it was interesting to investigate which type of epidermal keratinocytes modulate melanocyte proliferation and function most. Normal human epidermal melanocytes (HMel) were incubated with melanocyte-conditioned medium (M-CM) and low Ca2+ and high Ca2+ keratinocyte-conditioned medium (K-CM) obtained from the same skin source of melanocytes. The morphology, proliferation rate and melanin synthesis were evaluated at days 3, 6 and 12 of incubation. The results showed no evidence of major morphological changes in the epidermal melanocytes with any of the conditioned media, although marked dendrite formation was observed in coculture of melanocytes and differentiated keratinocytes. On the other hand, low Ca2+ K-CM induced a mild but statistically significant stimulation of melanocyte growth in a time-dependent manner. The significant percentage increase was evident on day 6 (124.6%, P < 0.05) and on day 12 (138.1%, P < 0.01) of incubation. In contrast, high Ca2+ K-CM showed no significant effect on melanocyte proliferation (P > 0.05). Both low Ca2+ and high Ca2+ K-CM stimulated melanin synthesis, although synthesis induced by low Ca2+ K-CM was higher than that of high Ca2+ K-CM. The significant percentage increase induced by low Ca2+ K-CM was evident on day 6 (117.9%, P < 0.05) and on day 12 (127.8%, P < 0.05) of incubation, whereas it was evident with high Ca2+ K-CM only on day 12 (119.7%, P < 0.05) of incubation. It is concluded from the above data that keratinocytes grown at a low Ca2+ level release factors that stimulate melanocyte proliferation as well as melanin synthesis, whereas keratinocytes grown at a high Ca2+ level release factors that only stimulate melanin synthesis. This may provide an explanation of the anatomical position of melanocytes and may play a part in the pigmentary changes following injury to epidermal cells.
