Streptomyces speibonae sp. nov., a novel streptomycete with blue substrate mycelium isolated from South African soil.

An actinomycete with blue substrate mycelium was isolated from a soil sample in Cape Town, South Africa, and designated strain PK-Blue(T). The colour of the substrate mycelium was not sensitive to changes in pH. The organism produced hairy spores in Spirales-type spore chains. Chemical taxonomy indicated that it belonged to the genus Streptomyces. Strain PK-Blue(T) produced no diffusible pigments other than melanin, grew at 45 degrees C, did not degrade adenine and exhibited no antibacterial activity against Enterococcus faecium, Escherichia coli or Pseudomonas aeruginosa. Analysis of its 16S rRNA gene sequence and the results of physiological tests showed that strain PK-Blue(T) (= DSM 41797(T) = ATCC BAA-411(T)) represents the type strain of a novel species of Streptomyces, for which the name Streptomyces speibonae sp. nov. is proposed.

Identifying phase-specific genes in the fungal pathogen Histoplasma capsulatum using a genomic shotgun microarray.

A fundamental feature of the fungal pathogen Histoplasma capsulatum is its ability to shift from a mycelial phase in the soil to a yeast phase in its human host. Each form plays a critical role in infection and disease, but little is understood about how these two morphologic phases are established and maintained. To identify phase-regulated genes of H. capsulatum, we carried out expression analyses by using a genomic shotgun microarray representing approximately one-third of the genome, and identified 500 clones that were differentially expressed. Genes induced in the mycelial phase included several involved in conidiation, cell polarity, and melanin production in other organisms. Genes induced in the yeast phase included several involved in sulfur metabolism, extending previous observations that sulfur metabolism influences morphology in H. capsulatum. Other genes with increased expression in the yeast phase were implicated in nutrient acquisition and cell cycle regulation. Unexpectedly, differential regulation of the site of transcript initiation was also observed in the two phases. These findings identify genes that may determine some of the major characteristics of the mycelial and yeast phases.

Effect of Radix Ginseng and Radix Trichosanthis on the melanogenesis.

Melanogenesis is a well known physiological response of human skin exposed to ultraviolet light, genetic reasons and other sources. In this study, we conducted to evaluate the effects of Radix Ginseng (RG) and Radix Trichosanthis (RT) on the melanogenesis in the B16 melanoma cells. The cells were treated for 48 h with RT at concentrations ranging from 1 to 50 microg/ml, RG at concentration of 10-1000 microg/ml, or RG at various doses (10-1000 microg/ml) with 25 microg/ml RT. Treatment with RT alone dose-dependently suppressed tyrosinase activity and melanin content compared with untreated control, and significantly inhibited cell proliferation. However, RG at various concentrations did not exhibit any significant change of them. Treatment with RT in the presence of various concentrations of RG suppressed tyrosinase activity and melanin content, similar to treatment with RT alone, but slightly increased cell proliferation. Furthermore, tyrosinase protein level was significantly decreased in treatment with 25 microg/ml RT alone and with a combination of 100 microg/ml RG. These results indicate that treatment with RG and RT significantly inhibits the melanogenesis in B16 cells, and raise the possibility that this combination may be effective in the whitening agent for the skin.

Due to the complexity , the biological process of hair growth is still a work in progress.
