Modulation of antioxidant enzymes p21WAF1 and p53 expression during proliferation and differentiation of human melanoma cell lines.

The activities of antioxidant enzymes, and the expression of p21(WAF1) and p53 proteins were studied at different times after subculture during proliferation and differentiation phases. Two human melanoma cell lines were used: IPC182, which is a non-differentiating cell line, and IGR221, which spontaneously differentiates at the end of the exponential growth phase, as evidenced by a marked increase of melanin content and tyrosinase activity. In the two cell lines, the slowing of proliferation coincided with an increase in the activity and amount of immunoreactive superoxide dismutases (SOD1 and SOD2), and a decrease of catalase and glutathione peroxidase activities, and of the glutathione content. The levels of p21WAF1 and p53 proteins were found to be lower in confluent than in proliferative cells. Several parameters were modified only during the differentiation phase of IGR221 cells; in these cells the increase of tyrosinase activity was highly correlated with the increase in SOD2, GST, glutathione reductase, and G6PD activities. The level of glutathione was found to be lower in differentiated IGR221 than in non-differentiated IPC182 cells. These results suggest that p21WAF1 and p53 proteins are not involved in the spontaneous differentiation process of melanoma cells, and that abnormal regulation of the cell cycle inhibition pathway occurred in these cells. The results sustain the hypothesis that alterations of antioxidant enzyme expression are involved in the control of proliferation and differentiation of melanoma cells. Alterations of SOD2 activity may be of particular importance, since variations are observed with both cell growth and cell differentiation.

Melanin in the extracellular matrix of germlings of Botrytis cinerea.

Previous work on the composition of the extracellular matrix of germlings of the plant pathogenic fungus Botrytis cinerea demonstrated the presence of carbohydrate, protein, and simple lipids; which, together, comprised 50-60% of the dry weight. Here we show that most of the remaining mass of the extracellular matrix consists of a chemically inert dark pigment with the electron paramagnetic resonance characteristics of a melanin. Scanning electron micrographs of the purified pigment, and transmission electron micrographs of thin sections made using the pigment indicate that it has a filamentous structure. We conclude that melanin is an important component of the extracellular matrix of germlings of B. cinerea. This is the first report of a melanin present in the extracellular matrix of a plant pathogenic fungus.

Effects of ultraviolet radiation on toad early life stages.

BACKGROUND: Exposure to harmful levels of ultraviolet-B radiation (UVB), a component of solar radiation, has been suggested as a potential cause of amphibian declines. METHODS: We measured solar radiation (UVB, ultraviolet-A, and visible) wavebands in breeding ponds of Bufo boreas (boreal toad, a montane species that has undergone severe population declines) and Bufo woodhousii (Woodhouse's toad, a plains toad that has not experienced declines)and examined tolerances of these species to simulated solar UVB exposures in the laboratory. RESULTS: We found larvae of both species to be tolerant of simulated solar UVB in excess of solar UVB levels observed in their breeding ponds. B. boreas tadpoles were more tolerant of simulated solar UVB exposure than B. woodhousii tadpoles, possibly because of greater amounts of photoprotective melanin in B. boreas skin. CONCLUSIONS: UVB levels observed in B. boreas habitats do not currently appear to constitute a threat to the survival of these animals; however, long-term (> 1 month) exposure to UVB levels comparable to levels associated with the water interface appears to reduce survival in B. woodhousii tadpoles. Therefore, future increases in surface and water column UVB radiation in bufonid habitats might pose significant survival risks to B. boreas or B. woodhousii populations.
