Variation in plasma steroids and reproductive traits in dimorphic males of corkwing wrasse (Symphodus melops L.).

Differences in plasma steroid levels may reflect behavioral, gonadal, or morphological specializations that characterize different male reproductive phenotypes in teleost fishes. In the present study, we investigated whether differences in plasma steroid levels exist between two distinct male morphs shown in the corkwing wrasse Symphodus melops. In addition, we examined differences in male reproductive traits, including gonad mass, sperm motility, and sperm concentration, between the two male types. Possible associations between plasma steroid levels and reproductive traits were also investigated. The results indicated that males with typical male secondary sexual characters (territorial males) had higher plasma levels of 11-ketotestosterone (11kT) compared to smaller males with female secondary sexual characters (female mimics). The female mimics in turn had higher plasma levels of both testosterone (T) and 17beta-estradiol. In addition, female mimics had relatively larger gonads and longer-lived sperm than territorial males. Relative gonad mass covaried significantly with the plasma levels of 11kT and T among the morphs, indicating that the relative gonad size correlates positively with the plasma level of these two steroids. However, there was no significant covariance between sperm traits and plasma steroids for the two male morphs. Hence, our results do not indicate any causal link between sperm quality and hormones. Inhibition kinetics of monoclonal antibodies against cytochromes P450.

Monoclonal antibodies (MAbs) inhibitory to individual cytochromes P450 (P450s) are of tremendous utility in identification of P450s responsible for the metabolism of a given drug or drug candidate in pharmaceuticals. In the present study, two inhibitory MAbs against CYP2D6 (MAb(2D6-50,) IgG(2b) and MAb(2D6-184), IgG(2a)) were developed by hybridoma technology to exhibit their high specificity and potency. The MAbs were further employed to assess the quantitative role (47-93%) of CYP2D6 to the metabolism of bufuralol in human liver microsomes from seven donors. Together with the MAb inhibitory to CYP3A4 as previously reported (Mei et al., 1999), the MAbs were used to study the inhibition kinetics of dextromethorphan O-demethylation (CYP2D6), testosterone 6beta-hydroxylation (CYP3A4) and aflatoxin B1 3-hydroxylation (CYP3A4), respectively, with an adequate size of sample measurement. A kinetic model was proposed to fit the experimental observations with three-dimensional nonlinear regression, thereby resulting in a solution of kinetic parameters, i.e., K(I), K(S), V(max), alpha, and beta (changes in K(I) or K(S) and V(max) in the presence of the MAb). As a result, dissociation constants (K(I)) of the MAbs for the enzymes and the maximal inhibition (beta) values for the P450-catalyzed reactions were predicted to have 0.04 to 0.25 microM and > or =94%, respectively. The results have demonstrated that the model can accurately predict the kinetic parameters and provide some insights into the understanding of the mechanism of MAb interaction with P450 enzyme in nature and the applications of the MAbs in qualitative and quantitative identification of P450s involved in drug metabolism.

Gender differences in the dietary lard-induced increase in blood pressure in rats.

We investigated the difference between male and female rats in the increase in blood pressure (BP) when they were fed a lard-enriched diet. We also investigated the effect of a gonadectomy, with or without testosterone treatment, on the dietary lard-induced increase in BP. Wistar-strain male or female rats were bilaterally castrated or ovariectomized. Some of them were implanted subcutaneously with silicon tubes containing crystalline testosterone. Each group was fed either chow alone or chow in which 50% of the energy content was from substituted lard. Systolic blood pressure (SBP) was determined weekly during each 7- or 11-week feeding period. A steady-state plasma glucose method was used to determine the insulin sensitivity. The dietary lard-induced increase in SBP was observed at 5 weeks after the start of the feeding in the sham-operated male rats. In the sham-operated female rats, SBP did not change from the basal values until 11 weeks into the experimental period. Castration eliminated the dietary lard-induced increase in SBP. Ovariectomy had no effect on SBP throughout the experimental period in both diet groups. In castrated males given testosterone, SBP increased in a dose-dependent manner. In ovariectomized females given testosterone, SBP increased significantly in the lard-enriched diet group. The dietary lard developed an insulin resistance in both the sham-operated and gonadectomized rats. However, SBP increased only in the sham-operated male rats. These results suggest that the dietary lard-induced increase in SBP depends on the presence of testosterone. The development of insulin resistance by dietary lard triggers the increase in SBP.

Thymic structural changes in relation to seasonal cycle and testosterone administration in wall lizard Hemidactylus flaviviridis (Ruppell).

Light microscopic and ultrastructural studies of thymus in wall lizard showed remarkable season dependent structural changes. In winter, the thymus was involuted and its cortico-medullary differentiation was not distinct. Thymocytes were sparsely distributed. The epithelial cells exhibited atrophic features such as an appreciable decrease in the nuclear-cytoplasmic ratio and accordingly reduction in cell organelles. The reconstruction of thymus commenced during spring and it became fully developed with marked delineation of cortico-medullary regions during summer. The thymus was then densely populated with thymocytes and epithelial cells showed voluminous cytoplasm having numerous cell organelles. The thymus regression started again by the beginning of autumn. The results suggest that the thymic development in wall lizard have inverse relationship with the androgen level, as the testicular steroidogenic activity was seen maximum during winter and least in summer. This assumption gets support by castration and testosterone replacement experiments. Castration of lizards during winter resulted in profound development of thymus with an appreciable increase in thymocytes mainly in the cortex region . The cortex became delineated from the medulla. Following testosterone treatment, the thymus underwent regression and was comparable to testis-intact lizard's thymus during winter season. After withdrawal of testosterone treatment, the thymus exhibited dense lymphoid and thymocyte population with a demarcation of cortico-medullary regions and sub-cortical region was regenerated.
