Opposing actions of adrenal androgens and glucocorticoids on alternative splicing of Slo potassium channels in bovine chromaffin cells.

Pituitary ablation (hypophysectomy) in rats was previously reported to cause a precipitous change in the relative abundance of two alternative splice variants of the "BK"- or "Maxi K"-encoding Slo gene in adrenal chromaffin cells. Inclusion of the optional "STREX" exon (STRess axis-regulated EXon) in a C-terminal splice site was reduced, in preference to the variant lacking an insert at this site. Adrenocorticotropic hormone (ACTH) injections prevented the drop in STREX inclusion, implicating stress-axis function, as opposed to other pituitary functions. Because ACTH promotes synthesis and release of glucocorticoids (corticosterone or cortisol, depending on species), we hypothesized that glucocorticoids applied directly would promote STREX inclusion. Contrary to predictions, we report that direct application of glucocorticoids to bovine cells in vitro decreased STREX inclusion. This effect was blocked by the glucocorticoid receptor antagonist RU38486. As with glucocorticoids, synthesis and release of the adrenal androgen dehydroepiandrosterone (DHEA) increases in response to stress or elevated ACTH levels in some species. We report that direct application of DHEA increased expression of the STREX variant in cultured bovine cells. Two other androgens, androstenedione and testosterone, had similar effects. We hypothesize that Slo splicing in adrenal chromaffin cells in vivo is differentially regulated by the integrative, concentration- and time-dependent actions of glucocorticoids and androgens, with potentially important ramifications for stress-evoked catecholamine secretion.

Hormone plasma levels from pituitary-gonadal axis in performance athletes after the 400 m run.

BACKGROUND: The aim of this study was to compare the concentration changes in hormones from pituitary - gonadal axis, induced by the 400 m run in the well-trained athletes (vice-champions in the Hall and Summer Athletic World Championship in 1999) to the changes observed in the competitors with shorter training period and achieving worse final results. METHODS: This research was conducted on 6 males - members of the Polish Olympic Team, who won vice-championship in the Hall and Summer World Championships 1999 and 6 athletes trained in the academic sport clubs. In the recent investigation, the 400 m run was assumed to be a stimulating impulse for evoking hormonal changes. The blood samples were taken from the elbow vein before the run, immediately after the effort and after the 24-hour rest. In the serum, the luteinizing hormone (LH), follicle-stimulating hormone (FSH), total testosterone (TT), free testosterone (FT) as well as the sex hormones-binding globulin (SHBG) concentrations were determined. RESULTS: During our research, immediately after the 400 m run in group I - the top class sportsmen - the statistically significant increase in both gonadotrophins (LH, FSH) was determined as well as the decrease in the total and free testosterone. In the group II - the athletes with the lower training level - the increase in FSH and the total and free testosterone concentrations was noticed. There were no statistically significant differences in the SHBG concentration. After-effort increase in the lactic acid concentration was observed in both groups. In the master group I, the increase in lactic acid concentration was higher than in group II. In both groups after the 24-hour restitution, the examined parameters, except LH levels in the group I, showed the concentrations similar to those before the effort. Analysis of the time needed to cover the distance of the race showed that the athletes from group I covered the distance of 400 m in the shorter time. CONCLUSIONS: The group of master class athletes, whose average intensive training period was 8 years, had higher VO(2max) and higher after-effort increase in the lactic acid concentration than in the group of sportsmen with the shorter training period (4 years), who had lower VO(2max), worse sport results and lower after-effort increase in the lactic acid concentration, gave different hormonal response (particularly TT, FT concentration) for the same exercise impulse. The difference based on the fact, that after the run in group I the decrease in the total and free testosterone levels and in group II the increase in the same parameters were observed. The observed hormonal changes in the master class athletes induced by the years-long anaerobic training might provide evidence for the reduction of functional reserves in gonads when compared to the group of less trained sportsmen.

The aqueous ethanol extract of Lygodii Spora (spore of Lygodium japonicum (Thunb.) Sw.) showed in vitro testosterone 5alpha-reductase inhibitory activity and in vivo anti-androgenic activity using growth of flank organ in castrated Syrian hamsters and hair regrowth after shaving in testosterone-treated C57Black/6CrSlc mice. From the lipophilic constituents of Lygodii Spora, oleic, linoleic and palmitic acids were identified as the main active principles inhibiting testosterone 5alpha-reductase.

Pharmacokinetics and pharmacodynamics of human chorionic gonadotropin (hCG) after rectal administration of hollow-type suppositories containing hCG.

To determine the effectiveness of human chorionic gonadotropin (hCG) administered rectally, we studied the pharmacokinetics and pharmacodynamics of hCG using a hollow-type suppository. HCG was not detected in plasma when only hCG was administered rectally, even at a higher dose (4,000 IU/kg body weight) than intravenous injection, because of its low bioavailability due to high molecular weight or degradation by proteolytic activity. To enhance the rectal absorption of hCG, the effectiveness of its coadministration with alpha-cyclodextrin (alpha-CyD), an absorption-enhancing agent, was investigated in male rabbits. HCG was detected in plasma following coadministration of hCG and alpha-CyD (10 mg/kg body weight) into the rectum. The plasma hCG concentration increased with increasing dose of alpha-CyD. The AUC(0-48) observed after coadministration of hCG and alpha-CyD at 30 mg/kg body weight was approximately four times higher than that of hCG and alpha-CyD at 10mg/kg body weight. HCG at a high concentration induced a rapid increase in the plasma testosterone concentration (74.2 +/- 3.4 ng/ml) 2 h after intravenous administration. However, the testosterone concentration 24 h after intravenous administration decreased to the physiological level (approximately 20 ng/ml) which had been observed before such administration. On the other hand, the maximum level of testosterone concentration (40.0 +/- 12.6 ng/ml) was observed 24 h after rectal administration of hCG (400 IU/kg body weight) in combination with alpha-CyD (30 mg/kg body weight). Moreover, the plasma testosterone concentration (31.0 +/- 11.4 ng/ml) obtained 72 h after rectal administration tended to be maintained at a higher level than that (14.4 +/- 0.9ng/ml) observed before the administration. These results suggest that the hollow-type suppository as a rectal delivery system of hCG is promising as a new mode of hCG therapy.

Hair loss is a problem in modern soceity. Examining the factors of hair growth may shed light on how hair loss might occur. How long can hair grow before it stops growing eventually if it does? Given that the hair growth rate is quite uniform and constant, somewhere between 0.3-0.5 millimeters per day, it's believed that the length of anagen, the growth phase, differs among individuals, and this is the major determinant to the maximum hair length. For some individuals, anagen may last ten years. Of course the length of the anagen is governed by genes, and the genetic background of the individuals. Non-genetic factors such as nutritional condition, weather, seasonal changes (hair may grow a bit faster during winter), taking medications, health condition may of course influence the rate of hair growth as well as hair loss. The shape of the hair, straight or curly, is dependent on the shape of the follicle. A circular or round hair follicle would generate straight hair, while the follicle with oval or elliptical shapes (in its cross-section) would produce a curly hair.
