Coordinated interaction of neurogenesis and angiogenesis in the adult songbird brain.

Neurogenesis proceeds throughout life in the higher vocal center (HVC) of the adult songbird neostriatum. Testosterone induces neuronal addition and endothelial division in HVC. We asked if testosterone-induced angiogenesis might contribute importantly to HVC neuronal recruitment. Testosterone upregulated both VEGF and its endothelial receptor, VEGF-R2/Quek1/KDR, in HVC. This yielded a burst in local HVC angiogenesis. FACS-isolated HVC endothelial cells produced BDNF in a testosterone-dependent manner. In vivo, HVC BDNF rose by the third week after testosterone, lagging by over a week the rise in VEGF and VEGF-R2. In situ hybridization revealed that much of this induced BDNF mRNA was endothelial. In vivo, both angiogenesis and neuronal addition to HVC were substantially diminished by inhibition of VEGF-R2 tyrosine kinase. These findings suggest a causal interaction between testosterone-induced angiogenesis and neurogenesis in the adult forebrain.

Changes in gene expression during aging in the Brown Norway rat epididymis.

Aging of the Brown Norway rat is associated with a decline in reproductive function. With advancing age, serum testosterone and sperm production decrease and sperm morphology is altered. In the epididymis, the site of sperm maturation and storage, dramatic histological and biochemical changes occur with age; these changes occur in a region-specific manner. In order to provide insight into the process of aging in the epididymis, we employed cDNA microarrays to analyze changes in gene expression with age in the initial segment, caput, corpus and cauda epididymidis. The overwhelming effect was a decrease in the relative intensity of gene expression during aging. In the initial segment, corpus and cauda epididymidis, more genes had decreased relative intensity with age than did not change. Interestingly, the magnitude of the decreases in relative intensity was considerably larger in the corpus and cauda epididymidis, where expression of 83% (211 of 254) and 62% (157 of 254), respectively, of the genes decreased by greater than 50% with age. This is in contrast to the initial segment, in which only 31% of genes (78 of 254) had relative intensities that decreased by at least 50%, and the caput epididymidis, the only segment where the largest proportion of genes did not change with age (less than 33% change between young and old). No genes had increased relative intensity with age throughout the tissue, however the expression of four transcripts was increased in a segment-specific manner. The expression of oxidative stress related genes, heat shock proteins and components of intracellular degradation pathways were examined in particular detail. We found that the relative intensity of some gene families decreased with age in a segment-specific manner; these decreases correlated well with the histological changes that occur in the aging epididymis.

Regulation of circulating soluble leptin receptor levels by gender, adiposity, sex steroids, and leptin: observational and interventional studies in humans.

Leptin is an adipocyte-secreted hormone important in energy homeostasis and diverse physiological processes. A circulating soluble form of the leptin receptor [soluble leptin receptor (sOB-R)] is the main leptin-binding protein and determinant of free leptin index (FLI), the presumed biologically active form of leptin. We performed observational and interventional studies to elucidate the regulation of sOB-R and FLI in humans. In a cross-sectional study (n = 118), leptin, gender, and adiposity were significant determinants of sOB-R. By multivariate analysis, estradiol (E2) and testosterone predict sOB-R, whereas insulin predicts leptin and FLI. In a frequent-sampling study (n = 6), sOB-R followed a significant circadian rhythm inverse to that of leptin, suggesting that leptin's biological activity may have an even more pronounced diurnal variation than originally thought. A 72-h fast in eight men decreased leptin levels by 80% and increased lymphocyte expression of leptin receptor mRNA and serum sOB-R levels by 100%. Physiological and pharmacological doses of recombinant-methionyl human leptin (rhLeptin) administered to fasted men prevented the fasting-induced increase of sOB-R levels, and pharmacological doses resulted in a decrease in sOB-R levels. These studies provide evidence that sOB-R is regulated by gender, adiposity, hormones, and rhLeptin administration. This may have important implications for the biological activity of leptin in disease states associated with abnormal leptin levels (e.g., obesity and anorexia nervosa).

Androgen-dependent expression of FcgammaRIIB2 by thyrocytes from patients with autoimmune Graves' disease: a possible molecular clue for sex dependence of autoimmune disease.

Thyrocyte expression of HLA class I and class II antigens and related accessory molecules would convert these epithelial cells into functional antigen-presenting cells. Here we show that whereas normal thyrocytes express FcRn, Graves' disease thyrocytes also express FcgammaRIIB2. We further find that expression of FcgammaRIIB2, but not FcRn, is repressed by dihydrotestosterone. By mediating the uptake and transport of autoantibodies, we suggest that these IgG Fc receptors contribute in various ways to the onset and/or progression of autoimmune thyroid diseases. The androgen-mediated decrease of FcgammaRIIB2 expression in Graves' disease thyrocytes also provides a rationale for the predominant susceptibility of women to develop an autoimmune thyroid disease. Our findings open up a new prospect to autoimmunity, linking the role of the target organ to the sex dependence in autoimmune disease.
