Saliva cortisol, testosterone and T/C ratio variations during a wrestling competition and during the post-competitive recovery period.

This study presents saliva cortisol (C) and testosterone (T) levels in a group of 15 young wrestlers (national and international) during a two-days competition. Values are compared to references established on a resting day (3 weeks before the competition). Post-competition recovery was studied by recording evening hormonal levels (5.30 pm) for 8 days. RESULTS: C levels increased sharply (about 2.5 fold resting levels) throughout the competition with no further changes in T levels. The rise in C appeared before entering the competition, traducing a striking phenomenon of anticipation. At the end of the competition, C levels fell very quickly (within 1.5 h) to basal value when T rose significantly, resulting in a very high T/C ratio. During the recovery period, C levels corresponded to basal ones and T remained high, resulting in a high T/C ratio (>30% than basal one) till the 5th day. CONCLUSION: Competition resulted in a low T/C ratio, considered as a catabolic phase. The recovery which is associated with a feeling of tiredness and an incapacity to train strenuously recognized by the trainers, paradoxically corresponded to a high T/C ratio (so-called anabolic phase).

Testosterone is a potential augmentor of antioxidant-induced apoptosis in human prostate cancer cells.

We have investigated the effect of antioxidant-induced apoptosis in human prostate cancer cell lines that is augmented by testosterone (T). In this study, DU-145 (androgen unresponsive), ALVA-101 (partially androgen responsive), and LNCaP (androgen responsive) were grown in tissue culture with RPMI 1640 medium, 5-10% fetal bovine serum (FBS), antibiotics and 5% CO2. Treatment with 2.5-20 microg/ml of PDTC significantly (P < 0.05, n = 6) lowered cell growth in all three cells 2-60% following treatment for 1-7 days. T (10(-12) M) alone enhances cell growth in androgen responsive cells. In contrast, the combination of PDTC and T significantly (P < 0.05, n = 6) augmented the PDTC induction of apoptosis in the androgen responsive cells, (ALVA-101 and LNCaP), but not in the androgen unresponsive cells (DU-145). PDTC reduced the nuclear NF-KB, as determined with an electrophoretic mobility shift assay (EMSA), to 50% of the control in LNCaP cells, 65% in ALVA-101 cells and 45% in DU-145 cells, but the combination of PDTC and T was not more potent than PDTC alone in any of the cell lines. PDTC suppressed both the AR mRNA and protein expression and reversed the stimulatory effect of T on androgen receptor (AR) protein synthesis in LNCaP and AVLA-101 cells. In conclusion, PDTC is a potent growth inhibitor and an inducer of apoptosis in human prostate cancer cells by reducing nuclear NF-kappaB and AR protein expression. PDTCs suppression of AR synthesis and nuclear NF-kappaB in response to T may contribute to its enhancement of apoptosis observed with T and PDTC compared to PDTC alone.

OBJECTIVE: Studies have reported the existence of marked sexual dimorphism in serum leptin levels in humans with women having approximately two to three times the levels of men. We have shown that this sexual dimorphism has a strong genetic component arising from a genotype by sex interaction, but adjusting leptin levels for testosterone eliminates this interaction. Because interactions such as genotype x sex can confound the detection of quantitative trait loci (QTLs), we wanted to determine if there are QTLs associated with the expression of leptin adjusted for testosterone. RESEARCH METHODS AND PROCEDURES: We performed a genome-wide scan using multipoint linkage analysis and implemented a general pedigree-based variance-component approach to identify genes with measurable effects on variation in leptin levels independent of testosterone in 318 Mexican Americans from the San Antonio Family Heart Study. RESULTS: We detected significant evidence of linkage (log of the odds ratio = 3.44) for a QTL on chromosome 22. DISCUSSION: Given these results, we hypothesize that a QTL on chromosome 22 may influence the level of leptin adjusted for testosterone.

Steroid Modulation of Muscarinic Cholinergic and alpha2-Adrenergic Receptor Density in the Nucleus Intercollicularis of the Japanese Quail.

Androgen modulation of neurotransmitter receptor density was investigated in the nucleus intercollicularis (ICo) of male and female Japanese quail (Coturnix coturnix japonica). ICo appears to play an important role in the neural control of testosterone (T) dependent vocal behaviour. Two receptor types were investigated in this nucleus; muscarinic cholinergic receptors, labelled using [3H]N-methyl scopolamine (NMS) as the ligand, and alpha2-adrenergic receptors, labelled using [3H]p-amino-clonidine (PAC) as the ligand. Changes in receptor density were assessed using in vitro quantitative autoradiography to ensure a high degree of anatomical specificity in the identification of any steroid effects. Gonadectomy was found to reduce the density of both [3H]PAC bindings sites and [3H]NMS in specific subregions of ICo. Gonadectomized animals treated with T had levels of receptor density similar to intact birds. However, the location within ICo of the effects of T was different for each ligand. [3H]NMS binding was modulated only in a rostral subregion of the nucleus while changes in [3H]PAC receptor density were found in the medial and lateral parts of ICo at a more caudal level. These changes in receptor density parallel changes in crowing frequency that are known to occur in males and females following castration and T treatment. The receptor systems may constitute a part of the neurochemical mechanism regulating vocal behaviour.
